Technology Overview

INT has developed a number of technical innovations to enable accurate field and point-of-care diagnostics. First, INT has developed an electronic sensor which can identify specific nucleic acid sequences. The electronic sensor provides the ability to screen for a wide variety of targets simultaneously. Second, to simplify use and improve field-ability, the company has developed a universal sample preparation process which can be fully automated. The new process uses improved sonication and magnetic separation techniques to clean and concentrate the DNA sample for identification. Finally, INT has developed a novel fluidic system to automate the entire process. The fluidic cartridge includes a unique rotary valve that minimizes consumable costs.

The process flow in the automated system is depicted below:

Sample Preparation

  • Lysis of cells and shearing of nucleic acids
  • Magnetic cleaning and concentration

PCR Amplification

  • 25 cycles of amplification
  • Oscillation between two temperature zones

Electronic Sensing

  • Hybridization to multiplexed sensor array
  • Metal development and electronic sensing

By automating the entire process from sample preparation to reporting of results, the system is easy to use and requires minimal training. The disposables contain all the required reagents and do not require refrigeration, eliminating the need for cold storage. The units are battery powered and will contain a wireless communications system and GPS, so that the system can collect time, location and test results automatically. Data will be collected wirelessly to enable real time disease surveillance.
